This study demonstrates that low-dose naltrexone (LDN) effectively restores bone material properties in high-fat diet (HFD)-induced type 2 diabetic mice by simultaneously modulating mineralization dynamics and collagen cross-linking integrity. Four experimental groups were analyzed: normal chow with saline (NC-Saline), normal chow with LDN (NC-LDN), HFD with saline (HFD-Saline), and HFD with LDN (HFD-LDN). Raman spectroscopy was performed on femoral periosteal surfaces to assess key compositional parameters. HFD-Saline mice exhibited a pronounced increase in mineral-to-matrix ratio, with v1 PO₄/proline at 21.66 ± 0.60, v1 PO₄/Amide III at 28.88 ± 0.60, and v1 PO₄/Amide I at 26.01 ± 0.94—significantly higher than NC-Saline controls (14.04 ± 0.59, 18.29 ± 0.44, 18.25 ± 0.38). Carbonate substitution was also elevated (carbonate/v1 PO₄: 0.193 ± 0.008 vs. 0.143 ± 0.004), indicating greater carbonate incorporation into the apatite lattice. Mineral crystallinity was reduced (1/FWHM: 0.0542 ± 0.0005 vs. 0.0548 ± 0.0004), suggesting disordered crystal growth. Collagen quality, assessed via the carbonate-to-amide I ratio, was significantly impaired in diabetic bones. AGE levels, measured through CML (1150 cm⁻¹) and PEN (1495 cm⁻¹) peak intensities normalized to CH₂ (~1450 cm⁻¹), were markedly increased in HFD-Saline mice, confirming glycation-mediated matrix damage.IL-13 Protein, HumanStorage & Stability In contrast, LDN treatment in HFD-fed mice brought all parameters toward normal values: mineral-to-matrix ratio decreased to 15.Fmoc-Thr(tBu)-OH supplier 45 ± 0.56, carbonate substitution dropped to 0.167 ± 0.008, crystallinity improved, and collagen quality recovered. AGE content was also significantly reduced. Nanoindentation of tibial cortical bone revealed that HFD-Saline mice had severely compromised mechanical properties—Young’s modulus at 32.PMID:34343231 04 ± 0.85 GPa and hardness at 1.32 ± 0.122 GPa—indicating brittle, weak bone. LDN treatment restored both properties to 37.47 ± 0.7 GPa and 1.70 ± 0.072 GPa, respectively, approaching control levels. Statistical analysis confirmed significant main effects of diet and treatment, with strong interaction effects across all variables. Post hoc tests showed that HFD-Saline differed significantly from NC-Saline, while HFD-LDN did not differ significantly from NC-Saline or NC-LDN. These findings indicate that LDN acts through dual mechanisms: it normalizes excessive and disordered mineralization while preventing pathological AGE accumulation and restoring functional collagen cross-links. This dual regulation preserves the hierarchical architecture of bone, enhancing its resistance to fracture. The results establish LDN as a potent agent for reversing diabetic bone fragility by targeting both mineral and organic matrix components, offering a promising therapeutic strategy for metabolic bone disease.MedChemExpress (MCE) offers a wide range of high-quality research chemicals and biochemicals (novel life-science reagents, reference compounds and natural compounds) for scientific use.
