**Background**
Cancer metastasis is a complex process involving the degradation of the extracellular matrix, which allows carcinoma cells to invade surrounding tissues and migrate to distant organs. A key player in this process is the urokinase-type plasminogen activator (uPA) system. uPA is a serine protease that converts plasminogen into plasmin, which in turn activates other proteases and degrades fibrin and other matrix components. Because of its critical role in promoting the invasive capacity of tumor cells, the uPA system has become a significant therapeutic target for preventing the spread of various malignancies. In this context, we will introduce a potent antimetastatic agent and uPA inhibitor – UKI-1.
**Definition**
UKI-1 (also known as WX-UK1) is a low molecular weight serine protease inhibitor that targets the urokinase-type plasminogen activator (uPA) with a Ki value of 0.41 μM.
**In Vitro and In Vivo Studies**
According to the UKI-1 description, this compound interferes with the plasminogen activation system at two distinct levels by inhibiting both the formation of plasmin directly and the activity of uPA. In terms of UKI-1 in vitro activity, treatment with concentrations ranging from 0.1 to 1.0 μg/mL has been shown to decrease tumor cell invasion by up to 50% in both the SCCHN line FaDu and the cervical carcinoma line HeLa. Furthermore, in invasion models utilizing highly invasive breast cancer and fibrosarcoma cells, UKI-1 effectively inhibits the migration of these cells through fibrin matrices.
Regarding UKI-1 In Vivo performance, the compound has demonstrated significant antimetastatic activities in animal models. Specifically, treatment with UKI-1 significantly reduces the number of metastatic lesions and inhibits overall tumor growth in rat models of mammary adenocarcinoma and pancreatic adenocarcinoma. Based on the UKI-1 technical information, the compound has a molecular weight of 613.81 and a chemical formula of C32H47N5O5S. In conclusion, UKI-1 is a potent uPA inhibitor that effectively suppresses the invasive and metastatic potential of various carcinoma cells.
